3D Bile-Duct Visualization and Quantification
Bile ducts located in the liver play a crucial role in the delivery of bile. Many liver diseases contribute to morphological abnormalities within the branching networks of liver bile ducts. Given this, visualizing abnormalities in liver bile duct morphology is crucial to designing robust therapeutic interventions.
The Full-Pipeline Liver Bile Duct service targets Cytokeratin 7 (CK7) throughout the intact liver. This service is well suited for studies investigating the complex branching networks of liver bile ducts. 3D visualization allows this service to provide comprehensive insights into morphological changes induced by genetic or pharmacological manipulations.

The Liver Bile Duct and Other Tubular Structures Quantification service utilizes Translucence Biosystems’ modified iDISCO+ tissue clearing protocol to render intact samples optically transparent. Staining for Cytokeratin 7 (CK7), found in liver bile ducts and Smooth Muscle Actin (SMA), a marker of vascular smooth muscle cells, facilitates the visualization of the bile ducts and vasculature across the intact liver.
Cleared, labeled samples are imaged on the Lightsheet Z.1 equipped with the Mesoscale Imaging System™. The Mesoscale Imaging System™ is optimized for imaging large intact tissues with unprecedented speed and resolution and is compatible with the high refractive index solutions used in tissue clearing protocols, such as iDISCO+.
Translucence’s imaging pipelines capture cellular-resolution, whole-sample images of liver bile ducts and other tubular structures, providing comprehensive insights into morphological changes induced by genetic or pharmacological manipulations.
We employ our in-house developed software solution, Stitchy, for automated light sheet image stitching. Stitchy is a robust software solution that stitches large imaging datasets 2.5-5.5 times faster than leading commercial software solutions. Stitchy enables seamless integration with downstream image analysis pipelines by eliminating intermediate file formats.
Translucence Biosystems labels and characterizes bile duct morphology, providing insights into liver pathologies and the impact of therapeutic interventions on bile duct architecture.
